Overview

A combination of **anetumab ravtansine** (a fully human anti-mesothelin IgG1 antibody-drug conjugate, conjugated to the maytansinoid tubulin inhibitor DM4) and **pegylated liposomal doxorubicin** (PLD, a liposome-encapsulated formulation of doxorubicin with polyethylene glycol for extended circulation). This combination is being studied for treatment of platinum-resistant epithelial ovarian cancer expressing mesothelin. Anetumab ravtansine targets the cell surface protein mesothelin, leading to delivery of cytotoxic DM4, while PLD adds cytotoxic chemotherapy with an improved pharmacokinetic profile over conventional doxorubicin. The approach leverages tumor-specific delivery and broad cytotoxic mechanisms. The combination has shown tolerability and promising clinical activity in phase Ib trials, justifying phase III evaluation in mesothelin-positive, platinum-resistant ovarian cancer patients[1][3][7].
