This combination regimen is a chemo-immunotherapy protocol consisting of the multi-target tyrosine kinase inhibitor anlotinib, the PD-1 checkpoint inhibitor toripalimab, and the standard-of-care chemotherapy doublet gemcitabine and cisplatin (GP). It is primarily being investigated as a first-line treatment for patients with recurrent or metastatic nasopharyngeal carcinoma (R/M NPC). The rationale behind this four-drug combination is to leverage the anti-angiogenic properties of anlotinib, which targets VEGFR, FGFR, and PDGFR, alongside the immune-potentiating effects of toripalimab to overcome the immunosuppressive tumor microenvironment. These targeted agents are combined with the cytotoxic effects of gemcitabine and cisplatin to maximize tumor cell death and improve clinical outcomes in advanced nasopharyngeal malignancies.
