Anlotinib + durvalumab is a combination therapy consisting of anlotinib, a novel oral multitarget tyrosine kinase inhibitor (TKI), and durvalumab, a monoclonal antibody that acts as a programmed death ligand-1 (PD-L1) checkpoint inhibitor. Anlotinib inhibits angiogenesis and tumor cell proliferation by targeting multiple receptor tyrosine kinases involved in tumor growth and vascularization. Durvalumab blocks the interaction between PD-L1 and its receptor PD-1, thereby enhancing anti-tumor immune responses. This combination has been investigated primarily for extensive-stage small cell lung cancer (ES-SCLC), both as part of first-line treatment regimens with chemotherapy and as maintenance therapy after initial chemotherapy response. Early-phase clinical studies suggest potential synergistic effects between these agents, leading to improved progression-free survival (PFS), objective response rate (ORR), and manageable safety profiles in ES-SCLC patients[1][2][3].
