Overview

This is a **combination regimen** of six drugs used in cancer therapy, primarily as a multimodal regimen for advanced or metastatic solid tumors (notably non-small cell lung cancer, NSCLC). The components are: - **Anlotinib**: an oral, small-molecule multi-target tyrosine kinase inhibitor with antiangiogenic and antiproliferative activity, inhibiting VEGFR-1/2/3, FGFR1-4, PDGFR-α/β, and c-Kit. Developed for the treatment of several solid tumors, approved in China for third-line NSCLC[1][2][6]. - **Erlotinib**: a small-molecule, oral EGFR tyrosine kinase inhibitor, indicated for NSCLC and pancreatic cancer. - **IBI308 (sintilimab)**: a humanized monoclonal antibody against PD-1, classified as a checkpoint inhibitor used in immunotherapy for various solid tumors[3][7]. - **Pemetrexed**: a multitargeted antifolate chemotherapeutic that inhibits thymidylate synthase, dihydrofolate reductase, and glycinamide ribonucleotide formyl transferase, used mainly in NSCLC and mesothelioma[4][8]. - **Gemcitabine**: a nucleoside analog chemotherapeutic that inhibits DNA synthesis, commonly used in lung, pancreatic, breast, and other cancers. - **Carboplatin**: a platinum-based chemotherapeutic causing DNA crosslinking and apoptosis, widely used in lung, ovarian, and other cancers. This multi-drug protocol aims to combine antiangiogenic, targeted, immunotherapeutic, and cytotoxic mechanisms to maximize anti-tumor efficacy. Such regimens are under investigation for first-line therapy of advanced NSCLC and other cancers[5][9].
