Overview

Anlotinib combined with immune checkpoint inhibitors (ICIs) is a therapeutic approach that combines two distinct mechanisms of action for cancer treatment. Anlotinib is a multi-targeted tyrosine kinase inhibitor that primarily blocks angiogenesis pathways by inhibiting VEGFR1/2/3, PDGFR α/β, FGFR1-4, and c-Kit. It works by suppressing tumor angiogenesis and interfering with various biological functions of malignant tumor cells. When combined with immune checkpoint inhibitors (such as PD-1/PD-L1 inhibitors), this approach demonstrates synergistic effects by enhancing immune cell infiltration, improving the tumor immune microenvironment, and providing dual action against tumor growth. Clinical studies have shown that this combination offers improved efficacy over monotherapy in treating various cancers, particularly non-small cell lung cancer (NSCLC), with manageable adverse events.
