Overview

Anlotinib + sintilimab is a combination therapy consisting of two distinct agents: anlotinib, a small molecule multi-targeted tyrosine kinase inhibitor with antiangiogenic and antiproliferative properties, and sintilimab, a fully human IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1). Anlotinib inhibits multiple receptor tyrosine kinases involved in tumor angiogenesis and growth, including VEGFR, FGFR, PDGFR, and c-Kit. Sintilimab blocks the interaction between PD-1 on T cells and its ligands PD-L1/PD-L2 on tumor cells or antigen-presenting cells, thereby enhancing antitumor immune responses. The combination has demonstrated synergistic antitumor activity in several solid tumors—including extensive-disease small cell lung cancer (ED-SCLC), non-small cell lung cancer (NSCLC), cervical cancer, endometrial cancer, microsatellite stable colorectal cancer (MSS CRC), liver cancer, and bile duct cancer—by combining immunotherapy with antiangiogenic effects[1][2][3][5][6][7][8].
