Overview

Anlotinib hydrochloride + irinotecan is a combination therapy consisting of anlotinib, a multi-targeted tyrosine kinase inhibitor, and irinotecan, a topoisomerase I inhibitor. Anlotinib inhibits angiogenesis and tumor cell proliferation by targeting multiple receptor tyrosine kinases including VEGFR, FGFR, PDGFR, and c-Kit. Irinotecan interferes with DNA replication in cancer cells by inhibiting topoisomerase I. This combination has been investigated primarily as a second-line treatment for advanced or metastatic cancers such as colorectal cancer (mCRC), esophageal squamous cell carcinoma (ESCC), and Ewing sarcoma after failure of first-line therapies. Clinical studies have shown that the regimen demonstrates promising anti-tumor activity with manageable safety profiles in these settings[3][4][6].
