Anthracycline + trastuzumab is a combination regimen used in the treatment of HER2-positive breast cancer. Anthracyclines (such as doxorubicin or epirubicin) are chemotherapeutic agents that intercalate with DNA, inhibit topoisomerase II, and disrupt DNA replication to induce cell death[7][10]. Trastuzumab is a monoclonal antibody targeting the HER2 receptor on cancer cells; it blocks HER2 signaling pathways essential for tumor growth and survival, induces antibody-dependent cellular cytotoxicity (ADCC), and may also affect cardiomyocyte function due to HER2’s role in cardiac repair[4][6][8]. The combination has been studied for safety in adjuvant settings for early breast cancer; while historically there were concerns about increased cardiotoxicity when used together, recent evidence suggests that concurrent use can be safe with careful cardiac monitoring[1].
