Overview

anti-ALPP CAR-T cells (TC-A101) is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy being developed by TCRCure Biopharma for the treatment of advanced solid tumors that express placental alkaline phosphatase (ALPP). ALPP is a tumor-associated antigen and glycosylphosphatidylinositol (GPI)-anchored protein that is frequently overexpressed in various malignancies, including ovarian, endometrial, and testicular cancers, while maintaining very low expression in healthy adult tissues. The therapy involves the genetic modification of a patient's own T cells using a retroviral vector to express a CAR specific for ALPP. Following lymphodepleting chemotherapy with fludarabine and cyclophosphamide, the engineered T cells are infused back into the patient to recognize and eliminate ALPP-positive tumor cells. It is currently being evaluated in Phase 1 clinical trials (such as NCT04627740) to assess safety, tolerability, and preliminary clinical efficacy.
