Overview

A combination cell therapy consisting of two distinct chimeric antigen receptor T cell (CAR-T) products—one targeting B-cell maturation antigen (BCMA) and the other targeting CD138 (Syndecan-1). Both BCMA and CD138 are highly expressed on malignant plasma cells in multiple myeloma. The rationale for combining these two therapies is to enhance antitumor efficacy by overcoming tumor heterogeneity and reducing the risk of antigen escape, a common mechanism of resistance in single-targeted therapies. Preclinical studies have shown that dual-targeting approaches can effectively eliminate malignant plasma cells while sparing non-malignant tissues. Early-phase clinical trials are ongoing or planned to evaluate safety and efficacy in relapsed or refractory multiple myeloma.
