Anti-BCMA CAR-T cells (specifically the HDS269B construct) is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy developed by The Second Affiliated Hospital of Henan University of Traditional Chinese Medicine. The therapy consists of a patient's own T cells engineered via gamma-retroviral transduction to express a second-generation CAR targeting the B-cell maturation antigen (BCMA), which is highly expressed on malignant plasma cells in multiple myeloma. The CAR construct includes a 4-1BB costimulatory domain to promote T-cell expansion and persistence, along with a truncated epidermal growth factor receptor (tEGFR) that functions as a safety switch for cell depletion if needed. It is currently being evaluated in Phase I clinical trials for the treatment of relapsed or refractory multiple myeloma.
