Overview

An investigational third-generation chimeric antigen receptor (CAR) T-cell therapy targeting B-cell maturation antigen (BCMA), developed by The First Affiliated Hospital of Soochow University in collaboration with Shanghai Unicar-Therapy Bio-medicine Technology. This cell-based immunotherapy is designed to treat multiple myeloma by utilizing a CAR construct that incorporates an anti-BCMA single-chain variable fragment (scFv) for antigen recognition, coupled with dual costimulatory domains (CD28 and OX40) and the CD3ζ T-cell activation domain. This third-generation design is intended to enhance the persistence and anti-tumor efficacy of the T-cells compared to earlier generations. In clinical trials, such as NCT03196414 and NCT03125577, this product is typically evaluated as part of a dual-target regimen, administered either sequentially or in combination with anti-CD19 CAR T-cells to address both mature plasma cells and potential progenitor cell populations in patients with relapsed/refractory or high-risk newly diagnosed multiple myeloma.
