The anti-BCMA immunotoxin (inactive mutant) is a recombinant immunotoxin (RIT) used as a negative control in preclinical research for the treatment of multiple myeloma. Developed by researchers at the National Cancer Institute (NCI), it consists of an antibody fragment (Fv or Fab) targeting B-cell maturation antigen (BCMA) fused to a truncated version of Pseudomonas exotoxin A (PE). The toxin domain contains a specific E553D mutation that completely abolishes its ADP-ribosylation activity, rendering the compound non-cytotoxic. It serves as a critical tool in scientific experiments to verify that the efficacy of active BCMA-targeting immunotoxins, such as LMB-70 and LMB-75, is derived from the delivery of a functional toxin rather than simple target binding or non-specific effects.
