anti-CD123 CAR-CIK (WT) is an experimental adoptive cellular immunotherapy consisting of cytokine-induced killer (CIK) cells engineered to express a third-generation chimeric antigen receptor (CAR) targeting CD123 (interleukin-3 receptor alpha). CD123 is a cell surface marker overexpressed in various hematological malignancies, particularly acute myeloid leukemia (AML) blasts and leukemic stem cells. The CAR construct features a single-chain fragment variable (scFv) derived from the 7G3 antibody clone, linked to CD28 and OX40 co-stimulatory domains and the CD3-zeta signaling moiety. Unlike traditional CAR-T therapies that often use viral vectors, these cells are generated using the non-viral Sleeping Beauty transposon system. The WT (wild-type) version utilizes the original high-affinity scFv, providing potent anti-leukemic activity, and serves as the baseline for comparison with affinity-tuned mutants designed to minimize on-target, off-tumor toxicity against healthy tissues like the endothelium and hematopoietic stem cells.
