Anti-CD19 CAR-T cells (CD28) refer to a class of chimeric antigen receptor (CAR) T-cell therapies engineered to target the CD19 antigen, a protein widely expressed on the surface of B cells. This specific architecture utilizes a CD28 co-stimulatory signaling domain alongside the CD3ζ primary activation domain. The inclusion of the CD28 domain is associated with rapid T-cell expansion and strong effector function, which is particularly effective in achieving high initial response rates in B-cell malignancies. Well-known FDA-approved products using this configuration include axicabtagene ciloleucel (Yescarta) and brexucabtagene autoleucel (Tecartus). These therapies are primarily indicated for various B-cell lymphomas and leukemias, and are increasingly being investigated for the treatment of B-cell mediated autoimmune diseases such as systemic lupus erythematosus (SLE).
