Overview

anti-CD19 CAR-T cells (Zhejiang University) is an autologous chimeric antigen receptor (CAR) T-cell therapy being developed by The Children's Hospital of Zhejiang University School of Medicine for the treatment of refractory systemic sclerosis. The therapy involves harvesting a patient's peripheral blood mononuclear cells, which are then transduced with a lentiviral vector to express a CAR targeting the CD19 antigen found on B cells. Following lymphodepletion with fludarabine and cyclophosphamide, the modified T cells are re-infused into the patient. The mechanism of action relies on the targeted elimination of CD19+ B cells, which play a critical role in the autoimmune pathogenesis of systemic sclerosis. It is currently being evaluated in a Phase 1 investigator-initiated clinical trial (NCT05085444) for both adult and childhood-onset refractory systemic sclerosis.
