Overview

The anti-CD274 F(ab) fragment is a monovalent antibody fragment derived from the papain digestion of the rat-anti-mouse CD274 (PD-L1) monoclonal antibody, specifically clone MIH6. As a checkpoint inhibitor, it targets Programmed cell death 1 ligand 1 (CD274/PD-L1) on the surface of tumor cells. By binding to CD274, the fragment competitively inhibits its interaction with the PD-1 receptor on T cells, thereby preventing the dephosphorylation of downstream signaling molecules like ZAP70 and PI3K. This blockade reverses the state of T-cell exhaustion—characterized by high expression of PD-1 and LAG3—and restores the proliferative and cytotoxic capacity of tumor-specific T cells. Preclinical studies in the TCL1 transgenic mouse model demonstrate that treatment with this fragment enhances the clearance of malignant B cells in vivo, suggesting its potential as a therapeutic strategy for chronic lymphocytic leukemia (CLL).
