Anti-CD3 + anti-CD28 refers to a combination of two monoclonal antibodies used to activate and expand T cells. This combination mimics the two-signal requirement for T cell activation: Signal 1 is provided by the anti-CD3 antibody, which binds to the CD3 epsilon chain of the T cell receptor (TCR) complex, and Signal 2 is provided by the anti-CD28 antibody, which binds to the CD28 co-stimulatory receptor. This dual signaling triggers robust T cell proliferation, cytokine production (such as IL-2), and effector function. While not a therapeutic drug itself, this combination is a critical research tool and a standard component in the ex vivo manufacturing of adoptive cell therapies, including Chimeric Antigen Receptor (CAR) T-cell therapies. It is also used in experimental models to study T cell-mediated pathologies, such as the collapse of immune privilege in hair follicles.
