Drug intelligence / Profile preview
Anti-CD3 F(ab')2 is a research-stage biologic consisting of the divalent F(ab')2 fragment of an anti-CD3 monoclonal antibody, with the Fc region removed. It targets the CD3 epsilon chain of the T-cell receptor (TCR) complex. Because it lacks the Fc domain, it does not bind to Fc receptors, thereby avoiding the massive T-cell activation, cytokine release syndrome, and associated morbidity/mortality typically observed with intact anti-CD3 antibodies. Anti-CD3 F(ab')2 has been shown to deplete pathogenic T cells, promote the expansion of Foxp3+ regulatory T cells (Tregs), and foster a TGF-beta-associated tolerogenic environment. It is widely studied in preclinical models of autoimmune diseases and transplantation, including type 1 diabetes, heart transplantation, graft-versus-host disease (GVHD), herpetic stromal keratitis, and hemophilia A.
