anti-CD30 CAR-T + PD-1 inhibitor is an investigational combination immunotherapy being developed by The Affiliated Hospital of Xuzhou Medical University for the treatment of CD30-positive lymphoproliferative diseases, including relapsed or refractory Hodgkin lymphoma and anaplastic large cell lymphoma. The therapy consists of autologous T-cells engineered to express a chimeric antigen receptor (CAR) that specifically recognizes the CD30 antigen (TNFRSF8), which is highly expressed on the surface of Reed-Sternberg cells and other lymphoma subtypes. To counteract the immunosuppressive tumor microenvironment and enhance the durability of the CAR-T cell response, a PD-1 inhibitor is administered concurrently or sequentially. This checkpoint blockade prevents the interaction between PD-1 on T-cells and its ligands (PD-L1/PD-L2) on tumor or stromal cells, thereby reducing T-cell exhaustion and improving clinical outcomes in patients who have failed standard therapies.
