**Anti-CD40 antibody + glucocorticoid** refers to an unnamed, preclinical antibody-glucocorticoid conjugate developed by Jiangsu Hengrui Pharmaceuticals. It combines a humanized, Fc-silenced antagonistic anti-CD40 antibody with a covalently linked glucocorticoid payload. The antibody component blocks CD40-CD40 ligand costimulation on B cells and dendritic cells, while CD40-mediated internalization delivers the glucocorticoid intracellularly to activate glucocorticoid receptor signaling. In reported preclinical studies, the conjugate inhibited CD40 ligand-driven immune-cell activation, suppressed T-cell-dependent antibody responses, and prolonged skin-allograft survival relative to unconjugated anti-CD40 antibody or systemic coadministration of glucocorticoid. The program was described as potentially applicable to autoimmune diseases and organ transplantation, including systemic lupus erythematosus.
