anti-CD47 F(ab')2 is a monoclonal antibody fragment consisting of two antigen-binding (Fab) regions linked by a disulfide bond, lacking the crystallizable fragment (Fc) region. It specifically targets CD47 (Cluster of Differentiation 47), a cell-surface protein often overexpressed on cancer cells that functions as a "don't eat me" signal by interacting with SIRPα on macrophages. By blocking this interaction, the F(ab')2 fragment neutralizes the inhibitory signal, thereby promoting the phagocytosis of malignant cells by the innate immune system. The use of a F(ab')2 fragment instead of a full IgG molecule allows for the blockade of the CD47-SIRPα axis without inducing Fc-mediated effector functions or certain toxicities, such as hemagglutination, which are sometimes associated with full-length anti-CD47 antibodies. In research settings, it is frequently investigated in combination with other tumor-targeting antibodies to synergistically enhance antibody-dependent cellular phagocytosis (ADCP).
