anti-CD5 CAR-NK cells is an investigational chimeric antigen receptor (CAR) natural killer (NK) cell therapy developed by Chongqing Precision Biotech Co., Ltd. for the treatment of relapsed or refractory T-cell hematologic malignancies, including T-cell acute lymphoblastic leukemia (T-ALL) and T-cell lymphoma. The therapy involves engineering NK cells to express a CAR targeting the CD5 antigen, a transmembrane glycoprotein constitutively expressed on the surface of most mature T-cells and malignant T-cells. Upon binding to CD5, the CAR-NK cells are activated to selectively eliminate CD5-positive tumor cells. This approach aims to overcome the limitations of CAR-T therapies in T-cell malignancies, such as the risk of "fratricide" (where CAR-T cells kill each other due to shared antigen expression) and the potential for contaminating the final product with malignant T-cells during manufacturing. CAR-NK cells may also offer a more favorable safety profile regarding cytokine release syndrome and graft-versus-host disease compared to traditional CAR-T therapies.
