The anti-CLEC2D + toll-like receptor 9 agonist conjugate is an immune stimulating antibody complex (ISAC) being developed by Immunitas Therapeutics. It consists of a fully human monoclonal antibody targeting CLEC2D (C-type lectin domain family 2 member D) conjugated to a CpG oligonucleotide, which serves as a Toll-like receptor 9 (TLR9) agonist. This therapeutic approach leverages CLEC2D-mediated internalization to deliver the CpG payload specifically into myeloid cells, B cells, and plasmacytoid dendritic cells. By triggering the TLR9 pathway locally within the tumor microenvironment, the conjugate stimulates inflammatory responses and enables the induction of sustained T cell and NK cell immunity. The program is currently in the research and preclinical stage for oncology indications, with proof-of-concept data presented at SITC 2023.
