This is a combination therapy consisting of an anti-CTLA4 (cytotoxic T-lymphocyte antigen 4) monoclonal antibody and high-dose interferon (HDI). Anti-CTLA4 monoclonal antibodies, such as ipilimumab or tremelimumab, are immune checkpoint inhibitors that block the CTLA4 receptor on T cells. This blockade enhances and prolongs T-cell activation by preventing inhibitory signaling through CTLA4, thereby promoting antitumor immune responses[1][2]. High-dose interferon (HDI), typically referring to high-dose interferon-alpha, is an immunomodulatory cytokine used in melanoma treatment to stimulate the immune system. The combination has been investigated in clinical trials for adjuvant therapy in melanoma patients at high risk of recurrence[3]. The rationale is that combining these two immunotherapies may synergistically enhance antitumor immunity.
