Overview

Regeneron's anti-GREM1 antibody is a fully human monoclonal antibody designed to neutralize Gremlin-1 (GREM1), a secreted protein that acts as a potent antagonist of Bone Morphogenetic Proteins (BMPs), specifically BMP2, BMP4, and BMP7. In healthy adults, GREM1 expression is typically low, but it becomes significantly upregulated in various pathological conditions, including fibrotic diseases of the liver, lungs, and kidneys, as well as in several types of cancer and osteoarthritis. By binding to GREM1, the antibody prevents it from sequestering BMPs, thereby restoring normal BMP signaling which promotes anti-fibrotic effects and proper cellular differentiation. Preclinical research has demonstrated that this antibody can reduce tumor burden in models of multiple myeloma and attenuate the progression of osteoarthritis and organ fibrosis.
