anti-HER-2001 + HBsAg is a novel Antibody-Vaccine Engineered Construct (AVEC) designed for the treatment of HER2-overexpressing cancers, specifically ovarian epithelial cancer. It consists of a synthetic anti-HER2 monoclonal antibody (anti-HER-2001) chemically conjugated to the Hepatitis B surface antigen (HBsAg) virus-like particle (VLP) using the bifunctional linker sulfo-m-maleimidobenzoyl-N-hydroxysuccinimide ester (SMBS). The construct operates through a therapeutic paradigm known as Redirected, Accelerated, and Amplified Prophylactic, HBV Vaccination-Induced Immunity Immunotherapy (RAAVIIT). By binding to HER2 receptors on tumor cells, the construct presents HBsAg on the cell surface, thereby redirecting the patient's existing, vaccine-induced anti-HBV immunity to the tumor site. This recruitment of polyclonal anti-HBsAg antibodies and immune effector cells (such as NK cells and cytotoxic T lymphocytes) triggers potent antibody-dependent cell cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC), leading to tumor cell apoptosis and necrosis while sparing healthy cells.
