Overview

This is a combination therapy consisting of an anti-interleukin-1 (anti-IL-1) antibody, interleukin-10 (IL-10), and interleukin-4 (IL-4). Each component targets different aspects of the immune response: * Anti–IL‑1 antibodies block the activity of IL‑1, a pro-inflammatory cytokine implicated in autoinflammatory and rheumatic diseases[2]. * Interleukin‑10 is a pleiotropic cytokine with strong immunosuppressive effects; it inhibits pro-inflammatory cytokine production by monocytes/macrophages and dendritic cells, suppresses antigen presentation, and limits T cell activation. It plays key roles in controlling inflammation in various diseases including colitis, neurodegenerative disorders, infections, and cancer[1][4]. * Interleukin‑4 is another anti-inflammatory cytokine that modulates immune responses by promoting Th2 differentiation and inhibiting Th1/Th17 pathways. The rationale for combining these agents is to achieve synergistic immunoregulatory effects. Studies have shown that combining IL‑4 and IL‑10 can provide greater chondroprotection, anti-inflammation, and analgesia than either alone or their simple combination—especially when delivered as a fusion protein—but even as separate agents they may counteract each other's potential adverse or proinflammatory effects[6][9]. The addition of an anti–IL‑1 antibody further suppresses inflammatory cascades driven by IL‑1. This combination has been investigated preclinically for conditions such as osteoarthritis (OA) where excessive inflammation leads to cartilage damage. In animal models of OA or arthritis, combinations involving these three components reduced cartilage pathology more effectively than single agents[6].
