This is a combination cellular immunotherapy approach that utilizes two engineered T cell technologies. The therapy combines anti-MUC1 CAR-T cells (chimeric antigen receptor T cells targeting the MUC1 antigen) with PD-1 knockout T cells (T cells with CRISPR-Cas9 mediated deletion of the programmed cell death protein 1 gene). PD-1 knockout enhances T cell functionality by preventing immune checkpoint inhibition, which normally attenuates T cell activation. Studies have shown that PD-1 knockout in T cells significantly enhances anti-tumor activity, increases cytokine secretion (TNF-α and IFN-γ), improves T cell motility, and maintains long-term persistence without impairing functionality or differentiation ability. This combination approach aims to overcome tumor immunosuppression mechanisms and enhance anti-cancer efficacy.
