anti-PD-1 antibody + mFOLFIRINOX is a combination chemo-immunotherapy regimen being evaluated for the treatment of metastatic pancreatic ductal adenocarcinoma (mPDAC). The regimen combines an anti-Programmed Cell Death Protein 1 (PD-1) monoclonal antibody, which serves as an immune checkpoint inhibitor to enhance T-cell activity against tumor cells, with the modified FOLFIRINOX (mFOLFIRINOX) chemotherapy backbone. mFOLFIRINOX consists of oxaliplatin (a DNA alkylating agent), irinotecan (a topoisomerase I inhibitor), and 5-fluorouracil (a thymidylate synthase inhibitor) modulated by leucovorin. This combination is primarily being developed and studied by Zhejiang University in randomized clinical trials to determine if the addition of immunotherapy can improve the poor prognosis associated with metastatic pancreatic cancer compared to standard-of-care chemotherapy alone.
