Overview

This combination therapy is a novel therapeutic strategy being evaluated by The Second Affiliated Hospital of Chongqing Medical University for the functional cure of chronic hepatitis B (CHB). The regimen involves the sequential or concurrent administration of peginterferon alfa (Peg-IFNα), nucleos(t)ide analogues (NAs), and an immune checkpoint inhibitor (anti-PD-1 antibody). The rationale behind this triple combination is to leverage the direct antiviral effects of NAs in suppressing HBV DNA, the immunomodulatory properties of Peg-IFNα, and the ability of PD-1/PD-L1 blockade to reinvigorate exhausted HBV-specific CD8+ T cells. The primary goal of this strategy is to achieve a functional cure, defined as sustained Hepatitis B surface antigen (HBsAg) loss, which is currently difficult to attain with standard monotherapies.
