anti-PD-L1 + IL-15 (also known by the research codes LH01 and LH05) is a research-stage immunocytokine fusion protein developed at Shanghai Jiao Tong University. It is designed to simultaneously block the programmed death-ligand 1 (PD-L1) immune checkpoint and deliver interleukin-15 (IL-15) mediated immune stimulation directly to the tumor microenvironment (TME). The prototype molecule, LH01, consists of an anti-PD-L1 antibody fused to an IL-15/IL-15Rα sushi domain complex, which elicits both innate and adaptive antitumor immune responses by activating natural killer (NK) cells and CD8+ T cells. A next-generation variant, LH05, was developed as a prodrug that masks IL-15 activity systemically and releases it selectively within the TME via tumor-associated protease cleavage, thereby reducing potential systemic toxicity while maintaining efficacy against 'cold' tumors.
