LIS1 is a glyco-optimized porcine polyclonal anti-human T-lymphocyte immunoglobulin (ATG) developed by Xenothera. Unlike traditional rabbit or equine ATGs, LIS1 is produced in genetically engineered pigs that lack the highly immunogenic porcine sugars alpha-1,3-galactose and N-glycolylneuraminic acid (Neu5Gc), which is intended to reduce the risk of serum sickness and neutralizing antibody formation in humans. The drug works by binding to a broad spectrum of T-cell surface receptors, including CD2, CD3, CD4, CD7, CD8, and HLA class I molecules, leading to rapid and sustained T-lymphocyte depletion through complement-dependent cytotoxicity (CDC), antibody-dependent cellular cytotoxicity (ADCC), and the induction of apoptosis. LIS1 is primarily being developed for the treatment of relapsed or refractory peripheral T-cell lymphoma (PTCL) and for the prevention of graft rejection in solid organ transplantation, such as kidney transplants.
