This is a combination therapy consisting of an anti-TIGIT monoclonal antibody, pomalidomide, and dexamethasone. Anti-TIGIT antibodies are immune checkpoint inhibitors that block the T cell immunoreceptor with Ig and ITIM domains (TIGIT) receptor on T cells and natural killer (NK) cells, thereby enhancing anti-tumor immune responses by preventing inhibitory signaling mediated by TIGIT's interaction with its ligands such as CD155[1][2][3][4]. Pomalidomide is an immunomodulatory small molecule that enhances T cell and NK cell function and has direct antitumor effects; it is approved for use in multiple myeloma[6][8]. Dexamethasone is a synthetic glucocorticoid corticosteroid used to reduce inflammation and modulate immune responses. The combination aims to synergistically enhance antitumor immunity—anti-TIGIT relieves immune suppression, pomalidomide stimulates immune effector cells, and dexamethasone manages inflammation or side effects. This regimen is primarily being investigated for hematologic malignancies such as multiple myeloma.
