A bispecific monoclonal antibody designed to simultaneously bind to the Vγ9 chain of the Vγ9Vδ2^+ γδ T cell receptor and to CD123, the alpha chain of the interleukin-3 receptor highly expressed in acute myeloid leukemia (AML) blasts. This engager redirects and activates Vγ9^+ γδ T cells (a specific subset of T cells) to selectively target and kill AML cells, aiming to overcome limitations and toxicities observed with conventional T cell redirection strategies. Anti-TRGV9 + anti-CD123 was developed by Janssen Biotherapeutics and has demonstrated selective γδ T cell activation, cytotoxicity against AML target cells in vitro and in vivo, and is in the preclinical development stage for AML treatment[1][3][4].
