Overview

**ANV419 + daratumumab** is a combination therapy under investigation for the treatment of relapsed/refractory multiple myeloma. ANV419 is an engineered, IL-2Rb-selective agonist—an antibody-IL-2 fusion protein designed to deliver high-dose interleukin-2 (IL-2) preferentially to natural killer (NK) and CD8+ cytotoxic T cells, thereby enhancing anti-tumor immunity. Daratumumab is a monoclonal antibody that selectively targets CD38, a glycoprotein highly expressed on myeloma cells, causing their death via mechanisms including antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), complement-dependent cytotoxicity (CDC), and direct apoptosis. Preclinical studies indicate that ANV419 increases NK cell activation and, when combined with daratumumab, significantly enhances NK cell-mediated killing of myeloma cells via ADCC. The combination is being evaluated in the OMNIA-2 Phase I/II clinical trial for relapsed/refractory multiple myeloma[1][3][5][7].
