anzutresgene autoleucel + mRNA-4203 (IMA203 + mRNA-4203) is an investigational combination immunotherapy being co-developed by Immatics and Moderna. The regimen pairs anzutresgene autoleucel (formerly IMA203), an autologous T-cell receptor engineered T-cell (TCR-T) therapy, with mRNA-4203, an mRNA-based cancer vaccine. Both components target the cancer-testis antigen PRAME (Preferentially Expressed Antigen in Melanoma). Anzutresgene autoleucel consists of the patient's own T cells engineered to recognize PRAME-derived peptides presented by HLA-A*02:01 on tumor cells. mRNA-4203 is a lipid nanoparticle (LNP)-encapsulated mRNA vaccine encoding PRAME, designed to act as a cell therapy enhancer by driving the activation and proliferation of the engineered TCR-T cells in vivo. The combination is being evaluated in a Phase 1a/b clinical trial for patients with HLA-A*02:01-positive, previously treated, unresectable or metastatic cutaneous melanoma or synovial sarcoma.
