Overview

AO-176 + bortezomib + dexamethasone is an investigational triple combination regimen for cancer, primarily under development for multiple myeloma. AO-176 is a humanized IgG2 monoclonal antibody targeting cell surface-expressed CD47, which acts as an anti-phagocytic signal, allowing tumor cells to evade immune clearance. Inhibition of CD47 by AO-176 promotes phagocytosis of tumor cells. Bortezomib is a small molecule proteasome inhibitor that disrupts proteasome-mediated protein degradation, leading to apoptosis in malignant plasma cells. Dexamethasone is a synthetic glucocorticoid steroid with anti-inflammatory and cytotoxic effects, frequently used in oncology regimens for its synergy with cytotoxic agents. The combination aims to enhance direct cancer cell cytotoxicity via bortezomib and dexamethasone while promoting immune-mediated clearance of tumor cells via AO-176-mediated CD47 blockade.
