AP-166 + Keytruda is a combination therapy comprising AP-166, an allogeneic natural killer (NK) cell therapy, and pembrolizumab (Keytruda), a programmed death receptor-1 (PD-1) blocking monoclonal antibody. Developed by Alphageneron, AP-166 utilizes NK cells that are specifically activated to target Heat Shock Protein 70 (Hsp70), a chaperone protein that is abnormally expressed on the surface of many solid tumor cells but not on healthy tissues. By combining these Hsp70-targeted NK cells with pembrolizumab's ability to prevent PD-1/PD-L1 mediated immune suppression, the treatment aims to overcome the immunosuppressive tumor microenvironment and enhance direct tumor cell lysis. The combination is currently being evaluated in Phase 2 clinical trials for the treatment of various solid tumors.
