Overview

The designation GD-Ruijin Hospital-osteosarcoma refers to a combination chemotherapy and targeted therapy regimen being evaluated in the Phase II PROACH trial (NCT03742193) at Ruijin Hospital, Shanghai Jiao Tong University School of Medicine. This regimen consists of the small molecule tyrosine kinase inhibitor apatinib combined with the chemotherapy drugs gemcitabine and docetaxel (GD). Apatinib selectively targets vascular endothelial growth factor receptor-2 (VEGFR-2) to inhibit tumor angiogenesis. Gemcitabine is a pyrimidine nucleoside analog that inhibits DNA synthesis, while docetaxel is a taxane that promotes microtubule assembly and stabilization, leading to cell cycle arrest. The regimen is specifically studied for the treatment of resectable pulmonary metastases in patients with osteosarcoma, typically administered as perioperative therapy followed by apatinib maintenance monotherapy.
