Overview

A combination therapy regimen comprising apatinib, an oral small-molecule tyrosine kinase inhibitor, and the IE chemotherapy backbone (ifosfamide and etoposide). Apatinib specifically targets vascular endothelial growth factor receptor-2 (VEGFR-2) to inhibit tumor angiogenesis. Ifosfamide is an alkylating agent that interferes with DNA replication, and etoposide is a topoisomerase II inhibitor that induces DNA strand breaks. This combination is primarily developed and studied by Peking University People's Hospital for patients with relapsed or refractory osteosarcoma who have failed first-line chemotherapy treatments. The regimen aims to overcome the short-lived activity of apatinib monotherapy and improve disease control in musculoskeletal lesions.
