**APG-115 + toripalimab** is a combination therapy under clinical investigation for advanced solid tumors, primarily advanced liposarcoma, malignant peripheral nerve sheath tumor, biliary-tract cancer, and adenoid cystic carcinoma. APG-115 (alrizomadlin) is a selective, orally administered small-molecule inhibitor of MDM2, designed to block the MDM2-p53 interaction and thereby activate the tumor-suppressor activity of p53. Toripalimab (JS001) is a humanized monoclonal antibody that inhibits the programmed cell death-1 (PD-1) receptor, promoting T-cell anti-tumor activity. The combination is intended to synergistically enhance anti-tumor immune responses and has shown promising safety and early efficacy signals in phase Ib/II trials[1][4][3].
