APG-2575 + homoharringtonine refers to a combination of APG-2575 (also known as Lisaftoclax), a selective BCL-2 inhibitor, and homoharringtonine (HHT), a plant alkaloid with established anti-leukemic properties. APG-2575 targets B-cell lymphoma-2 (BCL-2), promoting apoptosis in hematologic malignancies such as acute myeloid leukemia (AML). Homoharringtonine enhances anti-leukemic effect by promoting degradation of myeloid cell leukemia-1 (MCL-1), a protein that mediates resistance to BCL-2 inhibitors, via inhibition of the PI3K/AKT/GSK3β signaling pathway. The combination demonstrates synergistic anti-leukemic effects and is under preclinical and early clinical investigation for AML and other hematologic cancers[1][3][5].
