Aplitabart + FOLFIRI is an investigational combination therapy under evaluation for metastatic colorectal cancer and other solid tumors. Aplitabart (IGM-8444) is a monoclonal IgM antibody that acts as a Death Receptor 5 (DR5) agonist, promoting apoptosis in cancer cells. FOLFIRI is a standard chemotherapy regimen composed of folinic acid (leucovorin), fluorouracil (5-FU), and irinotecan hydrochloride, working synergistically to inhibit DNA synthesis and topoisomerase I, thereby suppressing tumor cell proliferation. Preclinical and early clinical data support the potential of the combination to provide robust anti-tumor responses, particularly when FOLFIRI upregulates DR5 expression and enhances aplitabart-mediated apoptosis. The combination is currently being tested in multi-agent clinical protocols, sometimes alongside bevacizumab, in patients with advanced or metastatic colorectal cancer[1][5][7].
