Drug intelligence / Profile preview
This drug is a **three-drug combination** therapy comprised of aponermin, pomalidomide, and dexamethasone. - **Aponermin** is a recombinant human tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) that induces apoptosis in tumor cells by activating death receptors DR4 and DR5[4]. - **Pomalidomide** is an immunomodulatory agent (IMiD) that inhibits myeloma cell growth, disrupts their microenvironment, induces cell cycle arrest, and promotes immune-mediated cell death[7]. - **Dexamethasone** is a synthetic glucocorticoid corticosteroid that exerts anti-inflammatory and cytotoxic activity, enhancing the efficacy of other anti-myeloma agents[7]. The combination is being investigated as a treatment for **relapsed or refractory multiple myeloma** (RRMM), particularly in patients who have received at least two prior lines of therapy, including lenalidomide and bortezomib. Clinical studies with related aponermin combinations (aponermin + thalidomide + dexamethasone) have shown improved progression-free and overall survival in this population with manageable side effects[4]. Combining pomalidomide and dexamethasone is already an established regimen for RRMM[6][7]. As of now, the specific triple combination of aponermin + pomalidomide + dexamethasone is under investigation.
