Overview

Apricoxib + erlotinib is an investigational combination therapy evaluated primarily for the treatment of non-small cell lung cancer (NSCLC) and pancreatic cancer. Apricoxib is a novel, selective cyclooxygenase-2 (COX-2) inhibitor that has been studied in phase II clinical trials. Erlotinib is a small molecule tyrosine kinase inhibitor targeting the epidermal growth factor receptor (EGFR), approved for use in NSCLC and pancreatic cancer under the brand name Tarceva. The rationale for combining these agents stems from evidence that COX-2 overexpression may promote resistance to EGFR inhibitors and contribute to tumor progression via mechanisms such as angiogenesis, evasion of apoptosis, and epithelial-to-mesenchymal transition. Clinical studies have explored this combination in biomarker-selected patients with advanced NSCLC who previously received platinum-based chemotherapy[4][7]. While some subgroups showed benefit, primary endpoints were not met overall.
