apricoxib + pemetrexed

Apricoxib + pemetrexed is an investigational combination therapy studied primarily for advanced non-small cell lung cancer (NSCLC). Apricoxib is a selective cyclooxygenase-2 (COX-2) inhibitor and nonsteroidal anti-inflammatory drug (NSAID), developed as an anticancer agent to potentially enhance the efficacy of standard chemotherapy. Pemetrexed is a folate antimetabolite that inhibits several key enzymes involved in nucleotide synthesis, leading to disruption of DNA and RNA synthesis in rapidly dividing tumor cells. The rationale for combining these agents was based on evidence that COX-2 overexpression correlates with poor outcomes in NSCLC and that COX-2 inhibition may sensitize tumors to chemotherapy. Clinical trials evaluated this combination versus standard regimens but did not demonstrate improved progression-free survival or overall benefit[1][3][5][6].
