Overview

APVAC1 + APVAC2 is a combination of two actively personalized peptide vaccines developed for the treatment of glioblastoma. The approach integrates two types of tumor antigens into standard care to maximize immune targeting in patients with newly diagnosed glioblastoma. - **APVAC1** consists of peptides derived from a premanufactured library representing unmutated tumor-associated antigens that are frequently overexpressed in glioblastoma tissue (shared antigens). For each patient, peptides best matching their tumor and immune profile are selected from this "peptide warehouse." - **APVAC2** comprises neoantigenic peptides unique to the patient's tumor and synthesized de novo based on individual tumor mutations identified through next-generation sequencing and immunopeptidome analysis. These mutated peptides are highly specific to each patient's cancer. Both vaccines are administered intradermally alongside immunomodulators such as GM-CSF and poly-ICLC (Hiltonol) to enhance immune responses. The combination aims to induce robust CD4+ and CD8+ T-cell responses against both shared and private tumor antigens. Clinical studies have shown high immunogenicity rates with favorable safety profiles[2][3][5].
