Overview

ARC-520 + pegylated interferon alpha-2a is an experimental combination therapy for chronic hepatitis B virus (HBV) infection. ARC-520 is a small interfering RNA (siRNA) therapeutic designed to induce RNA interference and selectively degrade hepatitis B viral mRNA transcripts, leading to decreased HBV antigen (especially HBsAg, HBeAg) and HBV DNA production[1][2][3][4][5]. Pegylated interferon alpha-2a is a long-acting form of interferon alpha, a cytokine with antiviral, immunomodulatory, and antiproliferative effects, used as a standard therapy for chronic HBV and hepatitis C. The combination was investigated to synergize direct antiviral activity (ARC-520) with immune system modulation (interferon), aiming for functional cure (loss of HBsAg). ARC-520 was developed by Arrowhead Pharmaceuticals. Clinical data showed ARC-520 significantly reduced HBsAg and HBeAg, but absolute reduction was moderate in some subgroups due to viral antigen production from integrated HBV DNA[1][3][4][5]. Pegylated interferon alpha-2a is developed and manufactured by Roche.
