ARC-521 + tenofovir is an investigational combination therapy studied for the treatment of chronic hepatitis B virus (HBV) infection. ARC-521, developed by Arrowhead Pharmaceuticals, is an RNA interference (RNAi) therapeutic designed to target HBV transcripts for degradation, reducing viral hepatic antigens. Tenofovir is a nucleotide reverse transcriptase inhibitor (NRTI) that suppresses HBV replication by inhibiting viral DNA synthesis. The combination leverages the rapid antigen-reducing capability of ARC-521 and the established antiviral activity of tenofovir, aiming for synergistic enhancement of HBV suppression. ARC-521 targets all genotype HBV transcripts, whereas tenofovir acts as a backbone agent for viral replication control. ARC-521 has shown activity in Phase 1/2 trials, and tenofovir is an approved standard-of-care NRTI for HBV and HIV[3].
